Novel targets are greatly needed to improve the treatment of chronic pain. We thus screened unstudied orphan receptors for a role in regulating pain. We found that two such orphans, GPR63 and GPR153, had such a role, while being nearly-unstudied themselves. Through our work, we have built a model whereby GPR63/153 act as negative feedback brakes on microglial activation, thus improving pain relief. We also report on the identification for the very first known drugs for these receptors.
